Nach 800 Jahren erwachen die Greed, Monster, die sich von der Gier der Menschen ernähren und eine Bedrohung darstellen. Der mittellose Reisende Eiji Hino wird eines Tages Zeuge, wie der junge Polizist Shingo Izumi von den Greed angegriffen wird. Als er zu seiner Rettung eilt, bekommt er vom Arm des Greed Ankh einen Gürtel und drei Medallien, um als Kamen Rider OOO gegen die Greed zu kämpfen. Um Eiji beistehen zu können, übernimmt Ankh kurzerhand den Körper von Shingo.
