Basierend auf dem gleichnamigen Gag-Manga, spielt »Polar Bear Cafe« in einer ganz normalen kleinen Stadt und handelt von den alltäglichen Vorkommnissen des Betriebs und vor allem von den Problemen und Erzählungen der Besucher, die gerne mal auf einen mehr oder weniger kurzen Abstecher vorbeikommen. Geführt wird das beliebte Café vom Eisbären Shirokuma, denn in dieser Stadt leben Tier und Mensch friedlich zusammen. Viele von ihnen sind im nahe gelegenen Zoo beschäftigt, der Tier und Mensch gleichermaßen eine Arbeitsstelle bietet, und so kann es sein, dass nach Feierabend die seltsamsten Gäste kurz mal reinschauen.

Zur Stammbelegschaft des Cafés zählen in erster Linie die Dauergäste Penguin und Panda sowie Sasako, die als Bedienung den Laden am Laufen hält. Gemeinsam unternehmen sie gelegentlich auch eigene Ausflüge und besuchen diverse Feste – gerne auch zusammen mit Grizzly, der nicht allzu weit entfernt eine Bar betreibt.
